Changes in antioxidant properties of pepper leaves (Capsicum annuum L.) upon UV radiation

Bell pepper (Capsicum annuum) is one of the most popular vegetables consumed worldwide. The leaves of pepper are rich in phenolics, including phenolic acids and flavonoids. These compounds are well known for their ultraviolet (UV) absorbing and antioxidant properties. While the change of the phenolic pattern is an intensive research subject, it is not yet well-known in pepper leaves, particularly in outdoor conditions. In this experiment, we examined the effect of UV radiation on the leaves of outdoor grown peppers, focusing on the UV-absorbing properties and antioxidant capacities. Three different total antioxidant capacity (TAC) measurements have been compared: (I) Folin-Ciocalteu Reactivity (FC), (II) Ferric Reducing Antioxidant Power (FRAP) and (III) Trolox Equivalent Antioxidant Capacity (TEAC). Moreover, non-enzymatic hydrogen peroxide scavenging antioxidant capacity was measured. Significant increase was detected only in FRAP, suggesting an elevation exclusively in the level of phenolic acids in case of UV exposed outdoor grown pepper leaves.

Evaluation of the content of polyphenols, flavonoids and tannins, the antioxidant capacity, and the antimicrobial activity of different organic and aqueous fractions of stems of Retama monosperma

Retama monosperma is an endemic plant of the Mediterranean region that has been traditionally used in folk medicine to treat various ailments. It contains a variety of bioactive phytochemicals and exhibited several biological activities. This study aimed to assess the phytochemical screening, total phenolic, total flavonoid, and total tannin compounds, as well as the antioxidant capacity and antimicrobial activity. The phytochemical screening involved color reactions, characteristic reagents, and precipitation methods. Total phenolic, flavonoid, and tannin compounds were quantified using colorimetric methods across four fractions. Antioxidant capacity was assessed using 2,2-Diphenyl-1-picrylhydrazyl radical scavenging, ferric reducing antioxidant power, 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging, and phosphomolebdenum assay. The antimicrobial activity was evaluated through disc diffusion method and the microdilution assay. Qualitative phytochemical tests revealed the presence of flavonoids, tannins, terpenoids, alkaloids, and sterols. Quantification of total phenolic, flavonoid, and tannin compounds confirmed the richness of polyphenolic compounds in all fractions. The antioxidant capacity measurements revealed that the ethanol fractions exhibited the highest antioxidant capacity in 2,2-Diphenyl-1-picrylhydrazyl, ferric reducing antioxidant power assays, and phosphomolebdenum assay. Conversely, the aqueous fraction showed highest activity in the 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) assay. Regarding antimicrobial activity, the ethyl acetate fraction demonstrated superior efficacy against Staphylococcus aureus, and Bacillus cereus. These findings suggest that R. monosperma could serve as a valuable source of antioxidant and anti-infective phytocompounds.

Assessing the impact of body composition, metabolic and oxidative stress parameters on insulin resistance as a prognostic marker for reactive hypoglycemia: a cross-sectional study in overweight, obese, and normal weight individuals.

Oxidative stress (OS) plays a pivotal role in the pathogenesis of insulin resistance (IR), particularly in its association with obesity. This study evaluate both the diagnostic and clinical significance of assessing oxidative status in patients affected by overweight and obesity displaying IR, especially with reactive hypoglycemic episodes (RH). A comprehensive examination of OS biomarkers was carried out, encompassing measurements of total oxidative capacity (TOC) and total antioxidant capacity (TAC). Our analysis results reveal noteworthy connections between OS levels and the severity of IR in overweight and obese patients. Moreover, in the study, we demonstrated the diagnostic utility of serum concentrations of TAC and TOC as indicators of the risk of RH, the occurrence of which, even at the stage of overweight, may be associated with increased OS and further development of obesity. Our findings imply that the evaluation of oxidative status could serve as a crucial diagnostic and prognostic tool for patients observed with IR and overweight and obesity. In conclusion, our study underscores the potential utility of assessing oxidative status in the context of IR and highlights the possibility of identifying novel therapeutic targets for the treatment of overweight and obese patients.

Colorimetric Sensing of Antioxidant Capacity via Auric Acid Reduction Coupled to ABTS Oxidation.

In this study, a simple and sensitive colorimetric assay has been developed for total antioxidant capacity measurement. The assay is based on the absorption measurement of the bluish-green oxidized product (ABTS·+) formed as a result of the oxidation reaction of the chromogenic reagent ABTS (2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) with gold(III). However, in the presence of antioxidants, the ABTS oxidation process is effectively suppressed due to the reduction of gold(III) ions to the zerovalent state forming gold nanoparticles (AuNPs). Relatively lighter colors and a significant decrease in absorbance are observed depending on the total antioxidant capacity. Taking advantage of this situation, qualitative and quantitative total antioxidant capacity (TAC) measurements, with the naked eye and UV-vis spectroscopy, respectively, could be successfully performed. The assay is named "auric reducing antioxidant capacity" (AuRAC) because the gold(III) ion-reducing ability of antioxidants is measured. The AuRAC assay was applied to dietary polyphenols, vitamin C, thiol-type antioxidants, and their synthetic mixtures. Trolox equivalent antioxidant capacity (TEAC) values obtained with the AuRAC assay were found to be compatible with those of the reference CUPRAC (cupric reducing antioxidant capacity) assay. The AuRAC assay was validated through linearity, additivity, precision, and recovery, demonstrating that the assay is reliable and robust. Compared to the simple TAC assays in the literature based on AuNP formation with subsequent surface plasmon resonance (SPR) absorbance measurement, this indirect assay has a smoother linear range starting from lower antioxidant concentrations. This method displays much higher molar absorption coefficients for antioxidant compounds than other conventional single electron transfer (SET) assays because 3-e- reduction of trivalent gold (i.e., Au(III) → Au(0)) produces three chromophore cation radicals (ABTS·+) of the assay reagent. The sensor has been successfully applied to complex matrices, such as tea infusions and pharmaceutical samples. The AuRAC assay stands out with its high molar absorptivity connected to enhanced sensitivity as well as its potential to convert into a paper-based colorimetric sensor.

Evaluating oxidative stress biomarkers in oncology nurses exposed to antineoplastic drugs: A cross-sectional study

PurposeAntineoplastic drugs (ADs) are widely used in cancer treatment. Nurses in chemotherapy centers are exposed to these drugs during preparation. They can affect healthy cells, leading to teratogenic and mutagenic effects, as well as oxidative stress. This study aimed to evaluate oxidative stress biomarkers in the nurses exposed to these drugs. MethodThis study was conducted on 30 nurses exposed to ADs and 30 nurses with no exposure to these drugs as non-exposed group. Oxidative stress biomarkers were measured in the blood serum samples of both groups, including malondialdehyde (MDA), catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx), total antioxidant capacity (TAC), and blood thiol groups. ResultsConsidering the possibility of confounding effect of nutritional supplement consumption, the effect of this factor was adjusted in the analysis. A significant difference was observed for CAT, SOD, thiol, and TAC biomarkers between two groups (P < 0.05). However, the difference in MDA and GPx biomarkers between two groups was not statistically significant. ConclusionsThe findings of the present study showed that supplement consumption has a significant effect on the biomarker of total antioxidant capacity. Thus, total antioxidant capacity measurement is advised as the best biomarker for tracking oxidative status in nurses exposed to ADs due to its capacity to measure all antioxidants in the body, except the thiol group, and its lower cost when compared to other biomarkers. Furthermore, it can be claimed that the consumption of nutritional supplements has a greater effect on the non-enzymatic biomarkers of oxidative stress than on enzymatic antioxidant system.

A preliminary study on a new approach for measurement of the antioxidant capacity of single molecules in food

Antioxidant activity refers to the rate of reaction between a specific antioxidant and a specific oxidant. On the other hand, antioxidant capacity is a measure of the quantity of a free radical in a concentration, neutralized by antioxidant molecules under examination. All common methods such as: Ferric Reducing Antioxidant Power (FRAP), Total Peroxyl Radical Antioxidant Parameter (TRAP), Oxygen Radical Absorption Capacity (ORAC), Crocin Bleaching Assay (CBA), 2,2-diphenyl-1-picrylhydrazyl (DPPH), and others have some technical limitations. All methods, before any analytical test, require a sample preparation, which is separated into hydrophilic and lipophilic fractions. However, this already represents an artifact, because some of the properties of the biological matrix are lost. The new OXEFIN unit defined as the ratio between the electrical power of the sample and the control allows us to compare the antioxidant capacities of single substances or a mixture of them, without the use of a target molecule and equipment such as a spectrophotometer or other optical detection instruments. Moreover, we are able to detect the total antioxidant capacity in an opaque solution without sample preparation(such as food). The milks range from 2.73 ± 0.02 to 0.20 ± 0.003 at 4 ml and 4.33 ± 0.04 to 0.26 ± 0.003 at 8 ml. The berries and fruit juices range from 5350 ± 38 to 673 ± 5.8 at 4 ml and from 5220 ± 40 to 915 ± 5 at 8 ml. The method proposed in this paper represents a new approach to the antioxidant capacity measurements reducing the cost, analysis time, the amount of reagents and avoid the possible chemical interferences during the radical chain reaction.

Umbelliprenin via increase in the MECP2 and attenuation of oxidative stress mitigates the autistic-like behaviors in mouse model of maternal separation stress.

Introduction: Autism spectrum disorder (ASD) is a complex neurodevelopmental condition. Maternal separation (MS) stress is an early-life stress factor associated with behaviors resembling Autism. Both MECP2 and oxidative stress are implicated in the pathophysiology of Autism. Umbelliprenin (UMB) is a coumarin compound with various pharmacological properties. Our study aimed to investigate the potential effects of UMB in mitigating autistic-like behaviors in a mouse model subjected to MS stress, focusing on probable alterations in MECP2 gene expression in the hippocampus. Methods: MS paradigm was performed, and mice were treated with saline or UMB. Behavioral tests consisting of the three-chamber test (evaluating social interaction), shuttle box (assessing passive avoidance memory), elevated plus-maze (measuring anxiety-like behaviors), and marble-burying test (evaluating repetitive behaviors) were conducted. Gene expression of MECP2 and measurements of total antioxidant capacity (TAC), nitrite level, and malondialdehyde (MDA) level were assessed in the hippocampus. Results: The findings demonstrated that MS-induced behaviors resembling Autism, accompanied by decreased MECP2 gene expression, elevated nitrite, MDA levels, and reduced TAC in the hippocampus. UMB mitigated these autistic-like behaviors induced by MS and attenuated the adverse effects of MS on oxidative stress and MECP2 gene expression in the hippocampus. Conclusion: In conclusion, UMB likely attenuated autistic-like behaviors caused by MS stress, probably, through the reduction of oxidative stress and an increase in MECP2 gene expression.

Supercritical Impregnation of Mangifera indica Leaves Extracts into Porous Conductive PLGA-PEDOT Scaffolds.

Plant leaves, such as those from Mangifera indica, represent a potential utilization of waste due to their richness in bioactive compounds. Supercritical CO2 allows these compounds to be incorporated into various matrices by impregnation. Combined with its ability to generate polymeric scaffolds, it represents an attractive strategy for the production of biomedical devices. For this purpose, conjugated polymeric scaffolds of biodegradable PLGA (poly(lactic-co-glycolic acid)) and PEDOT:PSS (poly(3,4-ethylenedioxythiophene)-poly(styrenesulfonate)), generated in situ by foaming, were employed for the supercritical impregnation of ethanolic mango leaves extract (MLE) in tissue engineering as a potential application. The extraction of MLE was performed by Enhanced Solvent Extraction. The effects of pressure (120-300 bar), temperature (35-55 °C), and depressurization rate (1-50 bar/min) on the physical/conductive properties and the impregnation of MLE were studied. The scaffolds have been characterized by liquid displacement, scanning electron microscope, resistance to conductivity techniques, measurements of impregnated load, antioxidant capacity and antimicrobial activity. Porosity values ranging 9-46% and conductivity values between 10-4-10-5 S/cm were obtained. High pressures, low temperatures and rapid depressurization favored the impregnation of bioactive compounds. Scaffolds with remarkable antioxidant activity were obtained (75.2-87.3% oxidation inhibition), demonstrating the ability to inhibit S. aureus bacterial growth (60.1 to 71.4%).

Effect of Inlet Air Temperature and Quinoa Starch/Gum Arabic Ratio on Nanoencapsulation of Bioactive Compounds from Andean Potato Cultivars by Spray-Drying

Nanoencapsulation of native potato bioactive compounds by spray-drying improves their stability and bioavailability. The joint effect of the inlet temperature and the ratio of the encapsulant (quinoa starch/gum arabic) on the properties of the nanocapsules is unknown. The purpose of this study was to determine the best conditions for the nanoencapsulation of these compounds. The effects of two inlet temperatures (96 and 116 °C) and two ratios of the encapsulant (15 and 25% w/v) were evaluated using a factorial design during the spray-drying of native potato phenolic extracts. During the study, measurements of phenolic compounds, flavonoids, anthocyanins, antioxidant capacity, and various physical and structural properties were carried out. Higher inlet temperatures increased bioactive compounds and antioxidant capacity. However, a higher concentration of the encapsulant caused the dilution of polyphenols and anthocyanins. Instrumental analyses confirmed the effective encapsulation of the nuclei in the wall materials. Both factors, inlet temperature, and the encapsulant ratio, reduced the nanocapsules’ humidity and water activity. Finally, the ideal conditions for the nanoencapsulation of native potato bioactive compounds were determined to be an inlet temperature of 116 °C and an encapsulant ratio of 15% w/v. The nanocapsules obtained show potential for application in the food industry.

The power of the QUENCHER method in measuring total antioxidant capacity of foods: Importance of interactions between different forms of antioxidants

Antioxidants play a crucial role in maintaining human health by counteracting oxidative stress and regulating redox balance within the body. The mixture of various antioxidant compounds in different forms (i.e., free, bound, insoluble) in food creates a redox active environment both in the human body and in the food system. Acting as both electron donors and acceptors while interacting with each other can either result in antagonism through pro-oxidative effects, or synergism through regeneration of one antioxidant by another. During the antioxidant capacity measurement, besides the individual antioxidant effects of the antioxidant components, these effects that occur because of their interaction with each other should be also considered. Classical antioxidant capacity measurement methods mostly concentrate on the fractions of foods that can be extracted with either water, alcohol, lipid, or acid/alkaline solutions. Antioxidants that cannot be extracted with any solvent are mostly ignored in these methods. On the other hand, the QUENCHER method, which allows direct measurement of antioxidant capacity foods without extraction, offers a rational solution to the limitations of traditional extraction-based methods. This approach considers the antioxidant capacity and interactions of all antioxidant forms that can be found in a food matrix, at the same time. This review provides detailed insights into the advantages of QUENCHER as a holistic approach for the accurate measurement of the antioxidant capacity of foods.

Anti-angiogenic and oxidative effects of sodium benzoate at different concentrations in chorioallantoic membrane model

&lt;b&gt;Aim:&lt;/b&gt; Due to the increased consumption of packaged foods, exposure to food additives is also increasing. Sodium benzoate (SB), a frequently used food additive, is generally used in alcoholic beverages, fruit, and vegetable juices, carbonated soft drinks, canned food, and various sauces. It is used to inhibit the formation of mold, yeast, and bacteria. This study was carried out to investigate the effects of SB exposure on angiogenesis and oxidant-antioxidant balance.&lt;br /&gt; &lt;b&gt;Materials &amp;amp; methods:&lt;/b&gt; Three different concentrations of SB, bevacizumab, and empty pellets were prepared, placed on chorioallantoic membrane (CAM), and examined for anti-angiogenesis. Total antioxidant capacity (TAC) and total oxidant capacity (TOC) measurements were made in the albumen samples, and oxidative stress index (OSI) value was calculated.&lt;br /&gt; &lt;b&gt;Results:&lt;/b&gt; The control group had no anti-angiogenic effect, but the bevacizumab group had a strong anti-angiogenic effect.10&lt;sup&gt;-3&lt;/sup&gt; M SB had a weak anti-angiogenic effect, but 10&lt;sup&gt;-4&lt;/sup&gt; M SB and 10&lt;sup&gt;-5&lt;/sup&gt; M SB showed no anti-angiogenic effect. TOC levels increased with SB in a dose-dependent manner. TAC levels decreased depending on the dose in the experimental groups with SB application. OSI levels increased depending on the dose increase in SB.&lt;br /&gt; &lt;b&gt;Conclusions:&lt;/b&gt; SB exposure caused a dose-dependent increase in oxidative stress and anti-angiogenic effect in CAM model.

Antioxidant Effects of Methanolic Extract of Quercus infectoria as a Supplement against Oxidative Stress Induced by Sub-acute Exposure to Arsenic; An in vivo Study

Background: Exposure to arsenic through drinking water is a global health problem that causes multisystem toxicity, mainly by inducing oxidative stress and impairing cellular energy. Objective: We aimed to evaluate the effect of Quercus infectoria gall extract (Qi) against oxidative stress induced by sub acute exposure to arsenic. Methods: The plant galls were extracted with methanol and were used for the determination of total phenolic content using Folin-Cio calteu reagent. Male Wistar rats were randomly divided into 8 groups of 6 animals and treated for 30 days. Negative and positive control groups received, respectively, normal saline and sodium arsenite (5.5 mg/kg) by gavage. Treatment groups received three doses of Qi (200, 400, and 600 mg/kg/day) by intraperitoneal injection 2 h. after oral administration of normal saline or sodium arsenite (5.5 mg/kg) (As-Qi). After 30 days, all animals were anesthetized with ketamine/xylasine and 2 mL of blood was taken for measurement of total antioxidant capacity (TAC) using ferric reducing antioxidant power (FRAP), lipid peroxidation (measurement of malondialdehyde (MDA)) and protein carbonylation of plasma. Results: Total phenolic content of the plant was determined to be 5.78 ± 0.23 mg gallic acid equivalent/g dried extract. The results of pharmacological studies indicated that in arsenic treated animals, a significant decrease in TAC, increase in lipid peroxidation and protein carbonylation happens compared to control group. Co-administration of Qi (600 mg/kg) with arsenic significantly increased TAC compared with arsenic group (0.245 ± 0.007 versus 0.183 ± 0.027 for arsenic) (p &lt; 0.05), while the serum MDA level (1.880 ± 0.499 versus 2.795 ± 0.112 for arsenic) and protein carbonylation were decreased in this group compared with arsenic treated animals (0.128 ± 0.007 versus 0.159 ± 0.009 for arsenic) (p &lt;0.01). In non-treated arsenic animals (NTAS), all three doses of Qi improved oxidative stress markers. Conclusion: Arsenic disrupt cellular antioxidant defense through overproduction of reactive oxygen species (ROS) and the Qi galls are able to revert some of these oxidant activities of arsenic. Previous studies have reported antioxidants in the plant and the present work can conclude that antioxidant effect of Qi is useful against happened oxidative stress in arsenic treated animals.

Effects of cooking on the composition of volatiles, total phenolic compounds, and antioxidant capacity of three Chilean seaweeds

Seaweed consumption is increasing around the world due to consumer demands for sustainable food sources, health benefits derived from bioactive compounds, and a source of novel flavors. Despite all the benefits that come with eating seaweed, consumers still resent of their sensorial properties. Seaweed is traditionally consumed raw or cooked, but there is still not enough information on how the sensory descriptors and bioactive compounds change when cooked. The aim of this study was to determine the change in total polyphenol content (TPC), antioxidant capacity (e.g., DPPH and ORAC values) and changes in volatile compounds of three Chilean seaweeds: Durvillaea antarctica, Pyropia spp. and Ulva lactuca when subjected to traditional cooking for 15 min at 100°C. In all three seaweeds, TPC decreased with cooking and so did the measured antioxidant capacity. Altogether, 46 volatile cpmpounds were identified in D. antarctica, 49 in Pyropia spp. and 47 in U. lactuca. The concentration of these volatile compounds was correlated with aroma sensory descriptors of the same samples. Consumer preferences may be attracted by the herbal notes of U. lactuca or the sweet, caramel, and umami flavors of D. antarctica and Pyropia.

The performance of hypochlorous acid modified Ag nanoparticle-based assay in the determination of total antioxidant capacity of Boswellia Serrata and Aronia

In this study, the SNPAC method was developed by using hypochlorous (ClO−) at pH 9.3. Polyvinylpyrrolidone (PVP) was used as a stabilizer. The existence of HOCl enables to formation stronger and more stable yellow emission which gives sensitivity. As a result, lower LOD and LOQ values were achieved by the AgNP–HOCl method. The obtaining lower values was attributed to the reoxidation of silver atoms with ClO− ions in the presence of PVP. The newly modified method was applied to the total antioxidant capacity (TAC) measurement of Boswellia Serrata, Aronia Melanocarpa tea, and juice. The results were compared with the results obtained by application of CUPRAC (Cupric ion Reducing Antioxidant Capacity) and FRAP (Ferric Reducing Antioxidant Power) assays. The TAC values of Boswellia Serrata, Aronia Melanocarpa tea, and juice were found as 6.18, 137.98, and 12.08 mmol TEAC/kg, respectively.

Brief research report pesticide occupational exposure leads to significant inflammatory changes in normal mammary breast tissue.

Studies have documented the high occurrence of several tumors, including female breast cancer, in populations occupationally exposed to pesticides worldwide. It is believed that in addition to direct DNA damage, other molecular alterations that indicate genomic instability are associated, such as epigenetic modifications and the production of inflammation mediators. The present study characterized the profile of inflammatory changes in the breast tissue of women without cancer occupationally exposed to pesticides. In samples of normal breast tissue collected during biopsy and evaluated as negative for cancer by a pathologist, oxidative stress levels were assessed as inflammatory markers through measurements of lipoperoxides and total antioxidant capacity of the sample (TRAP) by high-sensitivity chemiluminescence, as well as levels of nitric oxide (NOx) metabolites. The levels of inflammation-modulating transcription factors PPAR-γ (peroxisome proliferator-activated receptor gamma) and NF-κB (nuclear factor kappa B) were also quantified, in addition to the pro-inflammatory cytokines tumor necrosis factor-alpha (TNF-α) and interleukin 12 (IL-12). The levels of lipoperoxides, TRAP, and NOx were significantly lower in the exposed group. On the other hand, PPAR-γ levels were increased in the breast tissue of exposed women, with no variation in NF-κB. There was also a rise of TNF-α in exposed women samples without significant variations in IL-12 levels. These findings suggest an inflammatory signature of the breast tissue associated with pesticide exposure, which may trigger mechanisms related to mutations and breast carcinogenesis.

Assessment of the Antioxidant Capacity of Commercial Coffee Using Conventional Optical and Chromatographic Methods and an Innovative Electrochemical DNA-Based Biosensor.

As one of the most popular beverages in the world, coffee is a rich source of non-enzymatic bioactive compounds with antioxidant capacity. In this study, twelve commercial coffee beverages found in local Portuguese markets were assessed to determine their total phenolic and flavonoid contents, as well as their antioxidant capacity, by conventional optical procedures, namely, ferric reducing antioxidant power and DPPH-radical scavenging assay, and non-conventional procedures such as a homemade DNA-based biosensor against two reactive radicals: HO• and H2O2. The innovative DNA-based biosensor comprised an adenine-rich oligonucleotide adsorbed onto a carbon paste electrode. This method detects the different peak intensities generated by square-wave voltammetry based on the partial damage to the adenine layer adsorbed on the electrode surface by the free radicals in the presence/absence of antioxidants. The DNA-based biosensor against H2O2 presented a higher DNA layer protection compared with HO• in the presence of the reference gallic acid. Additionally, the phenolic profiles of the twelve coffee samples were assessed by HPLC-DAD, and the main contributors to the exhibited antioxidant capacity properties were caffeine, and chlorogenic, protocatechuic, neochlorogenic and gallic acids. The DNA-based sensor used provides reliable and fast measurements of antioxidant capacity, and is also cheap and easy to construct.

Study The Lung-Protective Effects of Riboflavin and Cyanocobalamin Against Lung Toxicity-Induced by Cyclophosphamide in Rats

Cyclophosphamide (CP) is a cytotoxic alkylating agent it's used associated with different side effects including lung toxicity. Vitamin B2 and vitamin B12 have lung-protective effects. This study was designed to evaluate lung-protective effects of both vitamins against lung toxicity induced by cyclophosphamide. seventy healthy adult albino male and female rats divided into seven groups each group containing ten rats were used in the present study and treated for seven days. Investigating the Content and Bioaccessibility of Phenolic Compounds In Roots of Rosa canina L. and Rosa pimpinellifolia L.

Rosehip is among the most important plants with high economic value, mainly used in foods and beverages from ancient times to the present. In this study, Rosa canina L. and Rosa pimpinellifolia L. roots, consumed as tea in Aktoprak Village of Erzurum province, were collected together with the fruits. The main goal of the study was to investigate the in vitro bioaccessibility of phenolic compounds in the roots and fruits of R. canina and R. pimpinellifolia by a simulated gastrointestinal digestion procedure. Methanolic and aqueous extracts were prepared for the analysis of phenolic compounds in roots, whereas only methanolic extracts were used for the analyses of fruits. Total phenolic and total flavonoid contents were evaluated spectrophotometrically, while four different methods were used for antioxidant capacity measurements. The quantification of individual phenolic acids, flavonoids, and anthocyanins was performed with HPLC-PDA. Results demonstrated that R. canina and R. pimpinellifolia have high levels of total phenolics, total flavonoids, and antioxidant capacity. The roots of R. pimpinellifolia and R. canina were observed to contain higher amounts of phenolics compared to the fruits. Epicatechin, 4-hydroxybenzoic acid, gallic acid, syringic acid, p-coumaric acid, naringenin, and ellagic acid were not determined in the fruit extracts of R. pimpinellifolia and R. canina, while they were detected in aqueous extracts of roots. Bioaccessibility analyses carried out on aqueous root extracts showed total phenolic recovery was 12.73% in R. canina, 10.71% in R. pimpinellifolia, and total flavonoid recovery was 0% in both species.

Stomatal Responses of Two Drought-Tolerant Barley Varieties with Different ROS Regulation Strategies under Drought Conditions.

Drought stress is a major obstacle to agricultural production. Stomata are central to efforts to improve photosynthesis and water use. They are targets for manipulation to improve both processes and the balance between them. An in-depth understanding of stomatal behavior and kinetics is important for improving photosynthesis and the WUE of crops. In this study, a drought stress pot experiment was performed, and a transcriptome analysis of the leaves of three contrasting, cultivated barley genotypes Lumley (Lum, drought-tolerant), Golden Promise (GP, drought-sensitive), and Tadmor (Tad, drought-tolerant), generated by high-throughput sequencing, were compared. Lum exhibited a different WUE at the leaf and whole-plant levels and had greater CO2 assimilation, with a higher gs under drought stress. Interestingly, Lum showed a slower stomatal closure in response to a light-dark transition and significant differences compared to Tad in stomatal response to the exogenous application of ABA, H2O2, and CaCl2. A transcriptome analysis revealed that 24 ROS-related genes were indeed involved in drought response regulation, and impaired ABA-induced ROS accumulation in Lum was identified using ROS and antioxidant capacity measurements. We conclude that different stomatal ROS responses affect stomatal closure in barley, demonstrating different drought regulation strategies. These results provide valuable insight into the physiological and molecular basis of stomatal behavior and drought tolerance in barley.

EFFECTS OF DIETARY NUCLEOTIDES SUPPLEMENTATION ON THE PERFOR-MANCE OF BROILER CHICKS

This study examined the effects of nucleotide supplementation on broiler chick growth performance, immunological response, carcass traits, meat quality, serum biochemical parameters, total antioxidant capacity, intestinal morphology, mortality rate, and economic efficiency measures of broiler chicks. A total of 180 one-day-old chicks (Ross 308) were distributed into 6 groups, each of which consisted of 30 chicks, and each group was divided into 3 replicates so that each replicate contained 10 chicks fed on six experimental diets as follows:  basal diet without oil or nucleotide (T1), basal diet with 1% soybean oil and without nucleotide(T2), 50 grams nucleotide per 100-kilogram diet with different fat sources (no oil(T3), 1% soybean oil(T4), 1% linseed oil(T5) &amp;1% fish oil(T6) respectively during the experimental period (5 weeks). Growth performance parameters were detected per stage period. Four chicks from each replicate were used at the termination of the experiment for analysis of the above mentioned measurements. Results revealed that supplementation of nucleotide in diets of broiler chicks improved feed conversion ratio, carcass traits, intestinal morphology, serum biochemical parameters, immunological response, bursa of Fabricius weight, and the best ratios were observed in groups fed nucleotide in combination with fish and linseed oil. Also, there was an increased economic efficiency in the SBO fed group(T2) then group fed nucleotides in mix with linseed oil (T5) and control (T1). In comparison to the control groups (T1 &amp; T2), groups fed nucleotides and PUFA oil sources had significantly lower n-6: n-3 ratio in breast muscle, and mortality rate.
